Abstract

2', 3'-didehydro-3'-deoxyithymidine (d4T) was obtained from 3',5'- anhydrothymidine (3) and potassium hydroxide in isopropanol. Compound 3 was prepared from the reaction of 3', 5'-di-O-mesylthymidine (2) with aqueous sodium hydroxide. 2 was synthesized from the reaction in ketone of thymidine (I) mesyl chloride and tertiary amines as N-methyl morpholine and 2,4-lutidine.
