Abstract

This review discuss synthetic procedures for obtainment of the 2',3'-didehydro-3'-deoxythymidine (d4T, Stavudine), that is used in the treatment of HIV infection. Different starting materials were used in these procedures such as 2'-O-deoxythymidine (thymidine), 5-methyluridine, and also thymine and ribose in total synthetic methods . In general these methods, for large scale, have serious difficulties due to: expensive and toxic solvents are used and many steps and prolonged reaction times are required. These drawbacks are superseded in only two reported industrial patents . In these reports thymidine and 5-methyluridine are used as row materials and 52 .5 and 60 % of d4T were obtained, respectively.
